SecureDoc from WinMagic (www.winmagic.com) does these things,
including options involving diskettes. It works very well. It's
not free.
Note that if you depend on a diskette in the way you describe,
sooner or later you will lose access to your computer when the
diskette fails or the data on it gets corrupted. Unless you make
a backup copy(ies)--in which case your hard disk security depends
on the physical security for one or more backup diskettes. That
is likely to be equivalent to far fewer bits than strong
encipherment. (How many key combinations are possible for the
lock? Is the lock the strong point in a weak door? Can you get
through the wall in 5 seconds with a sledge-hammer or a chain
saw? and so on.)
