File Rights

What's the best advisable way to set a directory structure.

Example:

D drive

Set the security on D and remove Everyone Full Control and then create
sub-folders under d drive and set the directory structure their.

Example:

D:\Users (set individual rights on sib-folder within here)
D:\Common
