The message :

is normal and required when changes are made to the configuration options..
I take it there was no other message in the errorlog?

Is this server a clustered sql server?
Is it a default or named instance?

If you look in the list of logins do you see the user that you are
currently registered as inSQL Enterprise Manager (SEM) ?
What SQL service pack are you running on the client where SEM is running?
If its not the same as the server, can you apply it?
Does it work now?

Could you try changing your registration method for SEM to use sa?

Does it work now?

If it does not could you run profiler and capture actions that are taking
place (TSQLStmt Starting and TSQL stmt Completed and all errors and
warnings, and ensure that you add in the error data column.
