Permissions on web server

Windows 2000 Server, IIS5, SP4.
Event Viewer displays following Warning:

The server was unable to add the virtual root '/' for the
directory 'D:\MyDir' due to the following error: Access is
denied. The data is the error code.

http://www.microsoft.com/contentredirect.asp.

D:\MyDir is the Default Web Site in IIS.
It runs a ISAPI application, in browser it works fine.

My question: what additional permissions should be set to
directory to get rid of Event Viewer warning?

at the moment I have:
Administrator - Full control,
Administrators - Full control,
Web Anonymous Users - Allow Read, Deny Write,
Web Appications - Allow Read, Deny Write,

I also tried to add System - Full Control, but this did
not remove Warning message from Event Viewer.

Second question is about removing default Sharing from
C:\ , C:\Winnt and D:\
I tried in Windows Explorer->Properties->Sharing->Do not
share this folder. But after restart the sharing is back.

under the chapter Removing Administrative Shares. It
says "In removing this registry entry, you need to make
sure that the Group Policy does not restore this entry
after you set it."
Which group policy? How and where can I change it to not
restore sharing?
