If you want anonymous user to have read / write
on the virtual directory, you need to ensure
iusr a/c at the physical directory itself has
related NTFS permission.

Use Windows Explorer, go to that folder, right mouse
properties, security tab, remove everyone add in
the user.

I won't recommend you to have anonymous one and
having write access. this is very risky, instead
you should create one user and assign such rights
and permissions to this users.
