Ping or Traceroute won't give any clues of port 25 availability.

Why don't you just ask your ISP?

If this host you're trying to reach is, by means of dns (most likely
hosted by your ISP) defined as mx for whatever domain, it wouldn't make
sense to block port 25.
If not, it wouldn't make sense to leave it open, because nobody will try
to send mail this direction.

Ahhm, besides spammers, of course, looking for a relay.
Maybe that's what you're testing?
