Well, it depends on which spy sweper are you depended on. Ad Aware from
Lavasoft and Spy Sweeper from Webroot do actually a really good job in
keeping my pc clean enough, and i do update them frequently to keep up
with new signatures.
Webroot Spy Sweeper has some shields that detect when you have a new
bookmark or some file is trying to redirect your home page of IE. It is
actually a good tool to be used, it even has the option to block any
software that tries to run when the next time boot happens.
You can try them for a trial period or a demo, and if you think it's
worthy, why not buying it? There is also the option of having host
files to trick your browser in thinking that the ad servers are in your
computer. Also Spy Bot Search and Destroy which is freeware can help
solve your problem.

Btw, using some time FireFox might help. I use actually both of them,
IE and FireFox. Firefox because i have installed some cool extensions
to it, one of them that blocks any advertisement based on Macromedia's
Flash, thus keeping my bandwith traffic down :). It just replace it
with a PLAY button in case i need to see it, which usually i don't
unless i trust the website.
So, consider switching totally to Firefox, try Webroot Spy Sweeper, Ad
Aware of Lavasoft and also Spybot Search and Destroy to fix your
problems. Remember to get offline when you do all this cleaning, and by
offline i mean no any single connection to any PC or Internet. Just
unplug the NIC cable, any modem and disable wireless if any.
Hope you can manage to do all this. If you need any host files, i
already have one with 1700 entries that block most of the ad servers.
